Abstract::
      Objectives Homocysteine thiolactone (HCTL) is a normal metabolite of amino acid metabolism,and has been proved as a potential radio-protecor. Its mechanism in radiation protection is unlcear yet.To understand its mechanism,the effect of HGTL on cell colony formation and the survival rate after irradiation, as well as on DNA double-strand break and rejoining were studied. Method Cell survival rate was estimated by colony formation test. DNA double-strand brealt and rejoining were measured by pulsed field gel electrophoresis. Results Ft was found that HCTL showed a mild protective effect on FTC cell survival after X-ray irradiation. Do increased from 1. 31 Gy to 1. 64 Gy. HCTL also stimulated colony formation under normal cell culture conditions;the plating efficiency was increased from 0. 23 to 0. 50 for FTC,from 0. 29 to fl. 35 for G361. It seemed to be caused by cell proliferation since the possi-bilty of alteration in cell attachment was excluded. But Ehere was no effect on CHO and fibroblast. Pos-sibly, the stimulation effect depends on cell type. ibroblasts do not response to HCTL. HCTL szimu-lated colony formation even when it was used after irradiation of cells,which deserves further attention for creating its usage for post-irradiation treatment. But HCTL did not show protective effect on DNA double-strand break induction nor repair. Conclusion HCTL stimulates cell proliferation under normal culture conditions and after irradiation. It implicates that the radio一protective effect may relate zo its physiological function rather than free radical scavenging.
