bjective To investigate the protective effect of TNF-α on hematopoietic impairment after radiation. Methods The radioprotective effect of TNF-α on Kunming mice whole body irradiated with lethal dose of 8.5Gy gamma rays was investigated. The effects of TNF-α on hematopoietic reconstitution of mice irradiated with sublethal dose of 7.0Gy gamma rays was studied with semisolid agar colony assay of CFU-GM. Using flow cytometry, the effect of TNF-α on the distribution of cell cycle of bone marrow in mice was analyzed. Results It was found that TNF-α could effectively raise 30 day survival rate after 8.5Gy irradiation and promote the recovery of WBC, BMC, CFU-GM and CFU-S when it was given before irradiation. Additionally, we found TNF-α could prevent the static hematopoietic progenitor populations from entering proliferative state. Conclusion TNF-α can promote hematopoietic impairment after radiation, which is an important mechanism of radioprotection. |
