| Objectives To study the relationships between genomic instability and leukemia lymphoma development, chromosome karyotypes of B10.Thy1.1 and STS/A mice at early periods after irradiation were analyzed. Methods G-banded chromosome karyotypes of B10.Thy 1.1 and STS/A strains from thymocytes of 12 to 33 days after leukemogenic fractionated whole-body X-irradiation were compared.Results Chromosome numerical and structural aberrations in B10.Thy1.1 mice were higher than those in STS/A mice.The average incidence of trisomy 15 was 37.5% from 12-32 days after irradiation in B10.Thy1.1 mice, but none was found in STS/A mice.Various kinds of chromosome translocations were observed and most frequent translocations occurred in E F region of chromosome 12.After thymocyte transplantation, 61.5% of lymphoma karyotypes in recipient mice were the same as in irradiated donor mice, most of them having trisomy 15 and translocations of chromosome 12. Conclusions Genetic instability expressed mainly as trisomy 15 and translocations of chromosome 12 plays an important role in neoplastic transformation of mouse lymphocytes. |
