A simple method of measuring DNA polymerase β-activity was established. This method is based on the following principle: DNA polymerases can enhance the ability of incorporating 3H-TTP into double-strand DNA gaps of activated DNA.In the condition of using selective inhibitor N-ethylmaleimide,which inhibits other DNA polymerases without effecting DNA polymerase β-activity,the incorporation rate of 3H-TTP in activated DNA can be considered as the reflection of DNA polymerase β-activity.Using this method,it was found that the β-activity of DNA polymerase in SMMC-LTNM hepatoma cells was two-fold higher than that in normal hepatocytes of nude BALB/cmice,and that the β-activity of DNA polymerase was not significantly affected by γ-rays within 30 Gy.This finding implies that the insensitivity to irradiation of some tumour cells might be related to their high DNA polymerase β-activity. |
