目的 对3例60Co辐照装置事故受照人员的外周血淋巴细胞亚群、血清蛋白组成及白细胞基因表达谱进行分析,寻找晚发性辐射损伤早期诊断治疗候选指标。方法 利用流式细胞仪检测淋巴细胞亚群百分比;琼脂糖凝胶电泳法分离和定量血清蛋白;基因芯片方法筛选辐射损伤差异表达基因,并用RT-PCR方法对差异表达基因进行验证。结果 与正常对照相比,患者在受照后3年其外周血CD4+T细胞持续降低,CD8+T细胞升高,CD4+T 细胞与CD8+T细胞比值降低;白蛋白含量下降,β球蛋白升高。基因芯片结果显示,上调差异基因285个,下调基因446个;RT-PCR对下调基因KDR,上调基因CEACAM8及OSM的验证结果与基因芯片检测结果一致。结论 CD4+T细胞及CD4+与CD8+T细胞的比值可作为晚发性损伤预测指标。 |

Objective To identify the candidate biomarkers for early diagnosis or the therapeutic targets in late ionizing radiation injury by analyzing the lymphocyte subgroups, serum protein levels and gene expression profiles in the peripheral blood of three60Co γ-ray accidentally-exposed patients during three years after irradiation. Methods Flow cytometry and agarose gel electrophoresis were used to investigate lymphocyte subgroups and serum protein levels, respectively. Gene expression profiles were obtained by using whole genome gene expression chip assay. Results Three-years after irradiation, the percentage of CD4+ T lymphocytes declined and the percentage of CD8+ T lymphocytes increased so that the ratio of CD4+ to CD8+ decreased, meanwhile, the serum albumin level declined whereas beta globulin increased in the patient peripheral blood. In addition, the expressions of 285 genes were up-regulated while those of 446 genes were down-regulated in the irradiated patient samples, where the expressions of KDR, CEACAM8 and OSM were further confirmed by real-time RT-PCR. Conclusions The changes of the percentage of CD4+ T cells and the ratio of CD4+/CD8+ T cells in peripheral blood may be useful as biomarkers of late radiation injury. |
