      目的 研究肿节风浸膏对羟自由基(OH·)的作用,并探讨其在豚鼠腮腺急性放射损伤中的作用。方法采用邻二氮菲-Fe2+氧化法,应用分光光度计检测不同浓度肿节风浸膏溶液对H2O2/Fe2+体系产生的OH·的清除作用;雄性豚鼠48只,随机分为对照组、照射组、药物组和药物+照射组,药物组和药物+照射组给予肿节风浸膏溶液,非药物组给予0.9%生理盐水,照射组和药物+照射组予γ射线局部照射头颈部,单次给量1500 cGy,照射后24 h取豚鼠腮腺组织,检测其中的总超氧化物歧化酶(SOD)活力和丙二醛(MDA)含量。结果肿节风浸膏溶液浓度分别为0.2、0.4、0.6和1.2 mg/ml时的OH·清除率分别为0.89%、17.52%、49.35%和89.89%,组间有量效关系。对照组、照射组、药物组和药物+照射组腮腺MDA含量分别为2.99、4.34、0.74、1.3 nmol/mgprot,组间比较差异有统计学意义(F=101.93,P=0.00);总SOD活力分别为249.6、223.02、262.23和165.05 U/mgprot,对照组、照射组和药物组的腮腺组织总SOD活力无差异,而药物+照射组显著低于其他3组。总SOD活力与MDA含量不存在线性关系(R=0.07)。结论肿节风浸膏在体外有显著清除OH·的作用;对γ射线诱导的氧化应激和腮腺急性放射损伤肿节风浸膏有潜在的保护作用,并可能对照射后腮腺组织总SOD活力有抑制作用。

      Objective To explore the effect of Sarcandra glabra extracts on hydroxy free radical (OH·), and acute radiation injury to guinea pig's parotid gland.Methods Spectrophotometry was used and 1,10-Phenanthroline-Fe2+ hydrocarbonylation was applied to observe the scavenging effect of Sarcandra glabra extracts on OH· in vitro. 48 male guinea pigs were randomly divided into four groups as control group, radiation group,medicine group which received Sarcandra glabra extracts but no radiation, medicine+radiation group which received extracts and radiation. The radiation group and medicine+radiation group were received a single dose of 15 Gy gamma-rays delivered to head and neck region. 24 hours after radiation parotid gland tissues were obtained, and total activities of superoxide dismutase(SOD) and malonaldehyde(MDA) content were measured.Results At the Sarcandra glabra extracts concentrations of 0.2, 0.4, 0.6 and 1.2 mg/ml in vitro, the scavenging rate of OH· were 0.89%, 17.52%, 49.35% and 89.89%, respectively. An obvious dose-effect relationship was observed with the concentration increased. MDA content of parotid gland were 2.99, 4.34, 0.74 and 1.3 nmol/mg of protein in control group, radiation group, medicine group and medicine+radiation group, respectively. There was a statistical significance among them(F=101.93,P=0.00). Total SOD activities of parotid gland were 249.6, 223.02, 262.23 and 165.05 U/mg of protein, respectively. There was no significant difference among control group, radiation group and medicine group, while medicine+radiation group was significantly lower than those of the above 3 groups. No linear relationship was observed between SOD and MDA(R=0.07).Conclusions Sarcandra glabra extracts could greatly scavenge OH· in vitro. It has a potential radioprotective effect against gamma-radiation-induced oxidative stress and acute injury to parotid gland. It might have an inhibitory action on total SOD activities of parotid gland after radiation.
