      目的 探索肠上皮内淋巴细胞(IEL)受照射后IFN-γ分泌的变化及其对肠上皮细胞增殖的影响。方法 采用ELISPOT法观察IEL受60Co γ射线8 Gy照射前后IFN-γ的分泌变化;利用新分离的IEL及大鼠隐窝空肠上皮细胞株(IEC-6)为离体细胞模型,通过混合、双室(隔离)培养系统,用MTT法观察IEL对IEC-6增殖活性的影响。结果 照射后IEL分泌IFN-γ增加,受照射IEL培养上清对IEC-6细胞增殖具有抑制作用,而双室培养可减轻抑制作用,加入抗IFN-γ抗体亦可减轻对IEC-6的抑制作用。结论 IEL受照后IFN-γ的分泌增加,对IEC-6的增殖具有抑制作用,IFN-γ是一种重要的介质因子,细胞与细胞紧密接触是IEL影响IEC-6增殖分化的一种重要方式。
英文摘要:
      Objective To explore the changes in IFN-γ secretion in intestinal intraepithelial lymphocytes (IEL) after radiation with 60Co γ-ray of 8 Gy and its influence on the proliferation of IEC-6. MethodsELISPOT was used to detect the changes of IFN-γ secretion in IEL after radiation. The influence of IEL on the proliferation of crypt cell line from rat jejunal epithelium (IEC-6) was analyzed by MTT assay, using the cell models of IEC-6 and incorporate_ and double_cell culture system.Results IFN-γ secretion in IEL after radiation was increased. And the culture supernatant of IEL after radiation had inhibitory action on the proliferation of IEC-6. The double-cell culture system could decrease this inhibitory action, and adding anti-IFN-γ have the same effect.Conclusions IFN-γ secretion in intestinal intraepithelial lymphocytes is enhanced after radiation and it has the inhibitory action on the the proliferation of IEC-6. Cell to cell contact may be an important way for IEL to inhibit the proliferation of IEC-6.
