目的 检测环氧合酶-2(COX-2) mRNA及蛋白在大鼠放射性口腔黏膜炎(ROM)组织中的表达水平,并探讨其在ROM中的作用机制。方法 建立放射性口腔黏膜炎SD大鼠模型,切取左、右侧颊黏膜,用半定量RT-PCR检测各标本COX-2的mRNA相对表达值,采用免疫组织化学SABC法检测COX-2蛋白在黏膜炎组织中的表达部位及表达强度。结果 成功地建立了SD大鼠放射性口腔黏膜炎动物模型。RT-PCR结果显示ROM黏膜组织能够表达COX-2 mRNA,而右侧自身对照组基本不表达COX-2 mRNA;免疫组织化学结果显示COX-2蛋白在ROM黏膜组织中广泛表达。结论 放射性口腔黏膜炎组织有COX-2 mRNA及蛋白的表达,与放射性口腔黏膜炎的严重程度有关。 |

Objective To detect the mRNA and protein levels of COX-2 in the tissues with radiation-induced oral mucosa(ROM) in rats and explore the mechanism of COX-2 in ROM. Methods The Sprague-Dawley (SD) rat model of ROM was established. Both sides of buccal mucosa were excised. The cDNA of COX-2 was synthesized using the semi-quantitative RT-PCR. The Streptavidin-Biotin-Complex Method (SABC Method) was employed to determine the localization, intensity and distribution of COX-2 protein expression. Results The SD rat model of ROM was established. The results of RT-PCR showed that mRNA of COX-2 could be expressed in the left buccal mucosa but rarely in the right buccal mucosa. Immunohistochemical analysis of COX-2 demonstrates that COX-2 protein can be expressed in the left side mucosa. Conclusions The mRNA and protein of COX-2 can be expressed in the mucosa of ROM. The expressions of COX-2 have correlation with the severity of ROM. |
