      目的 建立小鼠放射性肺损伤模型。方法 雌性C57BL/6小鼠90只,随机分作2组:(1)对照组(36只)不做任何处理;(2)照射组(54只)全肺单次照射12Gy。于照射后1、24、72h和1、2、4、8、16、24周,取照射组及相同鼠龄对照组小鼠肺组织,分别用HE、Masson染色在光镜下观察组织病理改变和胶原纤维沉积,免疫组织化学法来观察肺组织基底膜连续性和肺泡表面活性物质的表达。用碱水解法来测定羟脯氨酸含量。结果 与对照组比较,照射组小鼠的肺组织在照射后存在明显的病理组织学上的改变,早期以急性炎症反应为主,表现为肺充血、水肿、肺间质增厚,后期肺损伤以肺泡间隔的进行性纤维化为特征;免疫组织化学法的结果也进一步证实了病理组织学改变,与对照组相比,照射组小鼠的肺组织的基底膜连续性中断,并且肺泡表面活性物质的表达明显增多;同时,对照组湿肺羟脯氨酸含量为(0.763±0.029)μg/mg,照射组湿肺羟脯氨酸含量(0.875±0.009)μg/mg明显增高(P<0.05)。结论 该模型较好地模拟了放射性肺损伤的发生和发展过程,为今后进一步研究放射性肺损伤发生的机制提供了实验基础。

      Objective To establish a mice model of radiation-induced lung injury. Methods Ninety C57BL/6 mice were randomly classified into two groups, one received sham-irradiation (control group, Group C, n=36) and another irradiated thoraces with single fraction 12-gray X-rays (treatment group, Group X, n=54). The mice were sacrificed at 1, 24, 72 hours and 1, 2, 4, 8, 16, 24 weeks after the irradiation, lung tissues were extracted and processed for various analyses including HE and Masson staining, and immunohistochemistry. Hydroxyproline contents were also measured. Results Compared with Group C, significant histological and pathologic changes were observed and confirmed with immunohistochemistry in Group X. Low level of hydroxyproline (0.763±0.029) μg/mg was recorded in Group C, and an elevated level of hydroxyproline (0.875±0.009) μg/mg was found in Group X. The difference between the two groups were statistically significant (P<0.05). Conclusions A mice model of radiation-induced lung injury was established successfully.
