      目的 探讨meso-四(4-羟基苯基)卟啉[5,10,15,20-tetrakis(4-hydroxyphenyl)porphyrin]对电磁脉冲(EMP)照射离体人脐静脉内皮细胞的防护作用。方法 ①用MTT法进行meso-四卟啉对人脐静脉内皮细胞的毒性试验,找出最佳用药浓度,然后依此浓度作为防护药物浓度;②用化学发光法检测卟啉对人脐静脉内皮细胞在不同场强EMP照射下的防护效果。实验结果采用方差分析和独立样本t检验分析。结果 ①meso-四(4-羟基苯基)卟啉对人脐静脉内皮细胞的最佳用药浓度为0.01μg/ml;②经过EMP照射后,用药组化学发光强度(第1峰值和5s面积)明显低于对照组。结论 0.01μg/ml的meso-四(4-羟基苯基)卟啉对EMP具有一定的辐射防护作用。
英文摘要:
      Objective To study the protective effect of 5,10,15,20-tetrakis (4hydroxyphenyl) porphyrin (THP) on endothelial cell of vein-304 (ECV-304) exposed to electromagnetic pulse (EMP). Methods MTT method was used to determine the optimal concentration of THP toxicity reaction for ECV-304 cells, and then used this concentration in the protection test. Chemiluminescence was used to measure the radioprotective effect of THP on ECV-304 cells exposed to varying intensity of EMP at 100,200, or 400 kV/m. ANOVA and independent samples t-test were used to analyze the data. Results The optimal concentration of THP for ECV-304 cells was estimated to be 0.01 μg/ml.The chemiluminescence intensities of ECV-304 cells administered with THP were statistically significantly lower than those in the control group. Conclusion This study suggested that 5,10,15,20-tetrakis (4hydroxyphenyl) porphyrin at 0.01 μg/ml can exert radioprotective effect of certain extent.
