目的 探讨氨溴索对小鼠放射性肺损伤的保护作用。方法 采用雄性BALB c小鼠,体重25g左右,随机分为空白对照组(C)、单纯放射组(SI)和照射加氨溴索组(SI+A),每组80只,观察8个月。采用4MVX线直线加速器照射,射野2cm×2.5cm。常规照射,总吸收剂量为25Gy5次。氨溴索口服吸收50mg·kg-1·d-1,照射前1周服用至实验结束。照射后1~8个月取右肺组织进行羟脯胺酸含量测定,左肺组织用于病理检测,血浆进行转化生长因子β1(TGF-β1)含量测量。结果 SI组羟脯胺酸含量从第3个月开始增高,6月时最高,与7和8月基本一致。6月时,SI组明显高于SI+A组(P<0.01);SI+A组明显高于C组(P<0.01);TGF-β1含量从第2个月开始上升,3月达到高峰,4、5月开始下降,持续到8月。3个月时,SI组明显高于SI+A组(P<0.01);SI+A组高于C组(P<0.01)。结论 氨溴索通过羟脯胺酸和TGF-β1含量的改变减轻放射性肺损伤。 |

Objective To evaluate the protective effect of ambroxol on radiation induced pulmonary injury. Methods 240 BALB/c male mice were divided randomly into control group (C), irridiation alone (SI) and irridiation plus ambroxol (SI+A) groups. The right chests (size:2 cm×2.5 cm) of mice were irridiated by 4 MV linear accelerator for a total doses of 25 Gy,5 Gy/time,one time /day,5 time/week; ambroxol was orally administrered (50 mg·kg-1·d-1) from one week before irridiation until the mice were killed. Hydroxyproline content of lung-irridiated tissue and plasma TGF-β1 level were detected from 1 to 8 months. Results Hydroxyproline content increased starting from3 month, reaching peak at 5 and 6 month. and it was higher is SI group than that in A+SI group; contents in A+SI group was also higher than that of C group in 5-6 months respectively. Cell ultramicro-structure of lung tissue was damaged at different level in 1 and 3 months, a large fribric tissue was even accumulated at 6 months in SI group, while above cases were obviously slight in SI+A group.Conclusions Irridiation could induce the hydroxyproline increase in lung-irridiated tissue, change plasma TGF-β1 level lung tissue, ambroxol may reduce the radiation lung injury. |
