      目的 研究新的放射抗性肺癌D6-R细胞亚系在增殖活性、放射诱导的凋亡、DNA损伤与修复方面的特征,初步探讨D6-R细胞放射抗性的机理。方法 以D6-R和D6细胞为实验对象,MTT法测定细胞生长曲线,流式细胞仪和荧光显微镜分析细胞凋亡状况,碱性单细胞凝胶电泳检测DNA单链断裂与修复。结果 D6-R和D6细胞具有相同的增殖活性;照射前后均无明显的细胞凋亡发生;照射后D6-R、D6细胞的初始DNA单链断裂与修复能力存在统计学意义,放射抗性的D6-R细胞初始DNA单链断裂程度较轻,修复速率较快,修复后残存单链断裂较少。结论 初始DNA损伤程度较轻,修复能力较强可能是D6R细胞放射抗性的重要原因。
英文摘要:
      Objective To investigate the radioresistant mechanism of the newly established D6-R subline with respects to cell growth kinetics, radiation-induced apoptosis, DNA damage and repair. Methods D6-R and the parent D6 cells were engaged in this study. Cell growth kinetics was determined using MTT assay, apoptosis was analyzed by flow cytometry and fluorescence microscopy,and DNA damage and repair were detected using alkaline comet assay. Results D6-R and D6 cells showed the same growth kinetics pattern, and no evident apoptosis could be found pre- and post-irradiation in both cell lines. There were significant differences in radiation-induced initial DNA single strand breaks(SSB) and repair capacity between D6-R and D6 cells,i.e., D6-R showed less initial DNA SSB and better repair capacity. Conclusion The milder initial DNA damage and stronger repair capacity exhibited in D6-R cells might be the cause of their radioresistance.
