      目的 建立检测细胞外信号调节激酶2(ERK2)mRNA荧光定量的方法,了解ERK2在60Co诱发小鼠白血病中的作用。方法 利用60Co诱发BAIB/c小鼠发生白血病,建立辐射致癌动物模型,并按病理结果将动物分为3组:癌变组、辐射未癌变组和对照组。采用荧光定量RTPCR的方法,检测了ERK2在3组的mRNA表达情况,研究ERK2的mRNA表达与60Co诱发的小鼠白血病的关系。结果 癌变组骨髓细胞和胸腺细胞ERK2mRNA的表达均高于辐射未癌变组及对照组(P<0.05)。结论 建立了准确可靠的荧光定量检测ERK2mRNA的方法,ERK2可能参与60Co诱发的BAIB/c小鼠白血病的过程。

      Objective to establish a fluorogenic quantitative RTPCR(FQPCR) method for detecting the expression of ERK2 gene in tumor tissues and investigate the relationship between ERK2 mRNA expression and leukemia induced by γ-ray irradiation in mice. Methods After establishment of a leukemia model induced by γ-ray irradiation in mice,the animals were divided into three groups:the canceration group,the uncanceration group and the unirradiated group according to the pathological examination.Fluorogenic quantitative RTPCR(FQPCR) was set up to examine the expression of ERK2 in the tumor tissue of the leukemia induced by γ-ray irradiation in mice. Results The level of the ERK2 gene in the thymus and bone marrow cells of leukemic mice was significantly higher than in that of the control groups(P.<0.05). Conclusion The result of ERK2 gene expression detedcted with FQPCR is accurate and dependable.ERK2 may be involved in the canceration of irradiated mice.
