      目的 探讨辐射损伤后HR24L修复蛋白复合物的形成,为HR24L参与切除修复和重组修复提供生化证据。方法 利用质粒共转染和免疫共沉淀技术检测HR24L复合物中关键修复蛋白的存在及其在辐射损伤后的变化。结果 20Jm2紫外线照射损伤后2h,可检测到HR24L,复合物中存在有XPA。20Gyγ射线照射后2h,检测到Ku70参与HR24L复合物的形成,照射后8h可见Rad52存在。结论 辐射损伤后,切除修复蛋白和重组修复蛋白参与HR24L复合物形成的生化证据,进一步支持HR24L参与多种修复途径的论点。
英文摘要:
      Objective To investigate HR24L repair complex after DNA damage induced by radiation and provide biochemical evidence of the participation of HR24L in DNA excision repair and recombination repair. Methods Cotransfection and coimmunoprecipitations were employed to detect the presence of key repair proteins in HR24L compex. Results XPA was present in HR24L precipitations 2 h after 20 J/m2 UVirradiation.When cells were irradiated with 20 Gy γ-rays,Ku70 was precipitated with HR24L after 2 h and Rad52 was found in HR24L complex after 8 h. Conclusion By showing direct biochemical evidence for the presence of excision and recombination proteins in HR24L complex after radiation damage,the argument that HR24L participates in multiple repair pathway is reinforced.
