      目的 探讨神经酰胺和JNK信号通路在UVA诱导的细胞凋亡中的作用。方法 分别用MTT法和DNA梯形条带检测细胞死亡率和细胞凋亡;Western blot分析JNK信号通路的激活情况。结果 UVA照射后,正常的淋巴细胞JY出现明显的细胞凋亡,而MS1418细胞则没有。MS1418来自D型Niemann-Pick病人,酸性鞘磷脂水解酶遗传性缺陷,导致神经酰胺产生受阻。给予外源性神经酰胺,两种细胞都出现明显的细胞凋亡,表明UVA诱导的细胞凋亡是通过神经酰胺介导的。UVA激活了JY而不是MS1418细胞的JNK信号通路;给予外源性神经酰胺,两种细胞的JNK信号通路都被激活,说明神经酰胺位于JNK信号通路的上游。结论 UVA诱导的细胞凋亡依赖神经酰胺和JNK信号通路。
英文摘要:
      Objective To investigate the role of ceramide and JNK signaling pathway in UVA-induced apoptosis. Methods Cell death rate and apoptosis were examined by MTT assay and DNA ladder analysis respectively.Activation of JNK signaling pathway was analyzed by Western blot. Results UVA-induced apoptosis was markedly observed in lymphocyte JY,but not in MS 1418 cells from Niemann-Pick type D patients with an inherited deficiency of acid sphingomyelinase,resulting in inhibition of ceramide production.Once exogenous ceramide was administered,apoptosis occurred in both JY and MS 1418 cells,indicating UVA induced apoptosis via ceramide.UVA radiation activated JNK signaling pathway in JY but not in MS 1418, and exogenous ceramide stimulated JNK signaling pathway in both cells.The results indicated that ceramide was on the top of JNK signaling pathway. Conclusion UVA-induced apoptosis depends on ceramide and JNK signaling pathway.
