      目的 检测受照射后大鼠脑海马中氨基酸类神经递质的含量与变化。方法 一次全脑2、15和30Gy照射SD大鼠,照射后1周、1和3个月的SD大鼠经体脑微透析技术采集其海马组织的细胞外液,用高效液相色谱荧光法测定其中氨基酸类神经递质的含量。结果 30Gy照射后1周、1月和15Gy照射后1月3组大鼠谷氨酸和天冬氨酸的含量与对照组相比增加了1.6和2.5倍;而其他组兴奋性和抑制性氨基酸递质均没有明显改变。结论 大鼠大脑受照射后脑组织内兴奋性氨基酸神经递质有明显的增加,这为放射性脑损伤发病机理的研究提供了新的思路。
英文摘要:
      Objective To investigate the change in content of amino acid neurotransmitters in hippocampus of the irradiated rat brain,its relationship with radiation dose and the time course. Methods The whole brain of SD rats was irradiated with single dose of 5,15 or 30 Gy by 4 MeV electrons.The extra-cellular fluid in rat hippocampus was extracted by microdialysis in vivo,in which the contents of glutamic acid (Glu), aspartic acid(Asp).γ-aminobutyric acid(GABA),and glycine(Gly) were measured by high performance liquid chromatography, and the data were analyzed by the Student's test. Results The contens of amino acid neurotransmitters in un-irradiated and anaesthetized control rats were:Glu,289-317 μmol/L;Asp,47~54 μmol/L;GABA,85-113 μmol/L and Gly,185-203 μmol/L. By comparison to the control group,the contents of Glu and Asp increased significantly at one week and one month after 30 Gy irradiation,and one month after 15 Gy irradiation. But there were no obvious differences in amino acid concentration in all other groups. Conclusion The increase of Glu and Asp in the hippocampus of hight dose-irradiated rat brain suggests a possible association between the excitatory amino acid neurotransmitter hypothesis and the pathogenic process of radiation-injured brain.
