      目的 通过大鼠吸入氡及其子体后免疫器官的变化, 初步探讨氡及其子体对免疫系统的影响。方法 雄性SD大鼠吸入氡及其子体的累积剂量分别达66,111和174工作水平月(WLM)后, 收集支气管肺灌洗液(broncho alveo larlavage fluids,BALF),观察BALF中细胞计数和分类的变化。采用单细胞凝胶电泳(Single cellgelele ctrophor esis,SCGE)技术, 检测胸腺和脾脏细胞DNA链断裂情况。同时了解大鼠脾脏指数的改变。使用RT-PCR方法检测BALF细胞中IL6mRNA的表达情况。结果 大鼠吸入氡及其子体后, 各暴露组BALF中淋巴细胞比例与对照组相比明显减少, 而粒细胞增多(P<0.05).大鼠暴露达174WLM后胸腺和脾脏细胞DNA链断裂的迁移长度显着增加。111和174WLM暴露组的脾脏指数与对照组相比明显降低。174WLM暴露组BALF细胞中IL-6mRNA的相对表达量显着高于对照组(P<0.01),66和111WLM暴露组与对照组相比差异无显着性(P>0.05).结论 在本实验暴露剂量下, 氡及其子体可改变BALF中淋巴细胞和粒细胞的比例, 导致胸腺和脾脏细胞DNA的断裂损伤, 从而影响免疫器官的结构和功能。

      Objective To study adverse effects of inhaled radon and its progeny on immune system of rats. Methods Sprague-Dawley rats were exposed to radon and its progeny at cumulative doses up to 66,111 and 174 WLM (working level month),respectively.The numbers and types of cells in bronchoalveolar lavage fluids (BALF) were investigated.The spleen index was examined.The DNA migration in the thymus and spleen cells was detected with single cell gel electrophoresis(SCGE).RT-PCR was applied to determine the expression of IL-6 mRNA in BALF cells. Results It was shown that the proportion of lymphocyte of the exposed groups was significantly lower than that of the control group while the proportion of granulocyte was higher than that of the control group ( P< 0.05).The distance of DNA migration in the thymus and spleen cells increased in the 174WLM group.The spleen index of 111 WLM and 174 WLM groups decreased significantly compared with the control group.A high expression level of IL-6 mRNA in BALF cells was observed in the 174WLM group (P< 0.01)but there was no significant change in other two exposed groups compared with the control group. Conclusion The results suggest that in certain dose range radon and its progeny have adverse effect on structure and function of immune system of rats by affecting the proportion of lymphocyte and granulocyte in BALF, decreasing spleen index, and damaging the DNA of the thymus and spleen cells.
