目的 观察α骨化醇对肾辐射损伤骨质疏松大鼠骨质量的保护作用。方法 6月龄健康雄性SD大鼠30只,随机分为模型(Ⅱ),假手术(Ⅰ)和α骨化醇治疗(Ⅲ)3组,每组10只。以15Gy137Csγ射线照射大鼠肾脏局部制备肾辐射损伤模型,照射后1周,α骨化醇治疗组给予萌格旺0.1μg·kg体重-1·d-1,以1%羧甲基纤维素钠(CMC)制成混悬液灌胃,1ml/只,模型组以1%CMC灌胃,连续3个月。实验结束后,各组大鼠取L1~L4腰椎及右侧股骨,分别测定骨密度(BMD)、骨形态计量和生物力学等指标。结果 与Ⅰ组比较,Ⅱ组的腰椎骨密度明显降低,骨小梁体积、厚度减少,骨小梁间距增宽,矿化沉积率,股骨、腰椎的最大载荷明显降低(P<0.05,P<0.01)。α骨化醇治疗组与模型组比较,腰椎骨密度明显提高(P<0.01),骨小梁体积、宽度显著增加,骨小梁间距变小(P<0.05,P<0.01),矿化沉积率,股骨、腰椎的最大载荷明显提高(P<0.05,P<0.01)。结论 α骨化醇对肾局部辐射损伤导致骨质疏松大鼠骨质量有明显改善作用。 |

Objective To study the protective effect of alpha-calciferol on bone metabolism in rats with radiation injury to kidney. Methods Six-month-old male SD rats were randomly divided into three groups:model group, sham group, and alpha-calciferol (Bone-One) group, 10 rats each. The model group was developed by irradiation of rat kidneys with 15 Gy of Cs 137 γ-rays. One week after radiation, alpha-calciferol group was given orally Bone-One, 0. 1 μg·kgBW -1·d-1, suspended in 1% CMC, 1 ml a rat for 90 days. BMD of L 1-4, bone histomorphometry, bone biomechanical properties were measured in all rats at the end of experiment. Results BMD of L1-4 was significantly lowered, volume and thickness of trabecular bone were significantly decreased, space of trabecular bone increased, ineral apposition rate (MAR), maximal load of femur and lumbar vertebra decreased (P<0. 05, P <0. 01) in model group, compared with sham group. In alpha-calciferol group, BMD of L1-4, volume and thickness of trabecular bone increased, space of trabecular bone decreased significantly (P <0. 05, P <0. 01), MAR, maximal load of femur and lumbar vertebra increased significantly ( P <0. 05, P <0.01), compared with model group. Conclusion Alpha-calciferol improves disturbance of bone metabolism in rats with radiation injury to kidney. |
