| 目的 研究碱性成纤维细胞生长因子(bFGF)对辐射诱导的血管内皮细胞凋亡的作用及其信号转导途径。方法 以60Coγ射线照射建立原代分离培养的人脐带静脉内皮细胞凋亡模型,以流式细胞术(Annexin-V-FITC+PI标记)评价细胞凋亡率。观察不同浓度的bFGF和MEK的抑制剂U0126对细胞凋亡的影响。结果 bFGF对辐射诱导的脐静脉内皮细胞凋亡有明显的抑制作用,细胞凋亡率可从(35.22±5.5)%降至(11.67±3.8)%,而U0126可部分阻断bFGF的抗凋亡作用,细胞凋亡率由(10.59±1.6)%升至(17.96±1.4)%。结论 bFGF可能通过MAPK信号转导途径抑制辐射诱导的脐静脉内皮细胞凋亡。 |

| Objective To study the effect of bFGF on radiation-induced apoptosis in vascular endothelial cells and its signal transduction pathways. Methods Primary cultured of HUVEC (human umbilical vein endothelial cells) was irradiated with 60Coγ-rays to establish a cell apoptosis model. Flow cytometry with Annexin-V- FITC+ PI labeling was used to evaluate cell apoptosis. Different amounts of bFGF and MEK inhibitor U0126 were used to study their effects on radiation-induced endothelial cell apoptosis. Results We found that bFGF could inhibit radiation-induced apoptosis in HUVEC, i.e., the apoptotic rate decreased from (35.22±5.5)% to (11.67±3.8)%.Meanwhile, U0126 could partly block the anti-apoptosis effect of bFGF, the apoptotic rate increased from (10.59±1.6)% to (17.96±1.4)%. Conclusion MAPK signal transduction pathway may be involved in the inhibitory effect of bFGF on radiation-induced apoptosis in HUVEC. |
