      目的探讨川芎嗪对放射损伤小鼠骨髓造血微环境的影响及其信号传导机理。方法小鼠经60Coγ射线(6.0Gy)照射后即胃饲磷酸川芎嗪注射液,4mg/只,2次/d,连续13d.分别于照射后第3,7,14天处死小鼠,取尺骨制备石蜡切片,用免疫组化方法检测骨髓组织中因子Ⅷ相关抗原和VEGF受体flk-1的表达水平;培养骨髓基质细胞,用Westernblot方法分析骨髓基质细胞黏着斑激酶(磷酸化FAK)的表达水平。结果川芎嗪组小鼠造血恢复明显比对照组快,照射后第14天,川芎嗪组骨髓组织flk-1染色吸光度值(A)显着高于对照组(P<0.05),其基质细胞磷酸化FAK的表达在第14天时已接近正常水平,而对照组的表达仍较弱。结论川芎嗪可以促进骨髓内皮细胞上flk-1的表达和基质细胞中FAK的磷酸化,是改善放射损伤小鼠骨髓微环境、促进造血恢复的的机理之一。

      Objective To evaluate the effect of Ligustrazine on bone marrow hematopoietic microenvi-ronment and the mechanism of signal transduction in radiation-injured mice. Methods After irradiation by 6 0 Gy 60Co γ-ray, each mouse was given orally by 4 mg Ligustrazine twice a day for 13 days.On the 3th, 7th and 14th day, histopathologic sections from each paraffin-embeded ulnae were stained with anti-factor Ⅷ-related antigen and anti-flk-1.The femura were taken and the bone marrow nuclear cell suspension was made to culture bone marrow stromal cells(BMSCs), and the expression of phosphorylated FAK on BMSCs was assayed by Western blot. Results The recovery of hematopoiesis in the Ligustrazine-treated group was sooner than the control group.The expression levels of flk-1 and phosphorylated FAK in the Ligustrazine-treated group were significant higher than the control group( P<0 05). Conclusion The Ligustrazine can enhance the expression of flk-1 on bone marrow endothelial cells and the expression of phosphorylated FAK on BMSCs.This may be one of mechanisms of Ligustrazine to promote recovery of hematopoiesis after γ-irradiation.
