目的 探讨混合裂变产物致癌的分子机理和混合裂变产物内污染诱发免疫细胞的放射免疫毒理效应。方法 应用碱性单细胞凝胶电泳技术观察了混合裂变产物急性染毒时对大鼠外周血淋巴细胞DNA的损伤。结果 裂片核素内照射对大鼠外周血淋巴细胞DNA具有明显的损伤作用,慧星细胞率和DNA的损伤程度随染毒时间的延长、累积剂量增大而逐渐加重,且存在较好的剂量效应关系。结论 单细胞凝胶电泳技术检测的DNA单链断裂可作为毒物对健康影响的早期生物学指标。 |

Objective To explore the molecular biological mechanism of carcinogensis by mixed fission products. Methods Effects of DNA single-strand breaks of peripheral blood lymphocytes in rats induced by internal exposure with mixed fission products were analyzed by alkaline single cell gel electrophoresis assay. Results It was found that fission-debris nuclides caused increase of percentage of cells with migrated DNA, and DNA migration length in the lymphocytes showed a dose-dependent manner. Conclusion The results suggest that comet assay can be applied to monitor DNA single-strand breaks induced by internal exposure. |
