目的 观察电离辐射对小鼠腹腔巨噬细胞IL-12和NF-κB活性的影响以及两者间相互作用的关系。方法 采用酶联免疫吸附实验(ELISA)检测IL-12蛋白水平变化,通过电泳凝胶移动变化分析(EMSA)检测NF-κB结合活性的变化。结果 ①ELISA检测结果显示,腹腔巨噬细胞分泌的IL-12在0.075GyX射线全身照射后4~8h升高(P<0.05),2Gy照射后从2h开始升高,一直持续至48h仍高于假照组的水平(P<0.05),而且2Gy引起IL-12升高的幅度大于0.075Gy;②EMSA检测结果显示,NF-κB呈现两条特异带分别代表P65/P50、P50/P50两种二聚体,0.075Gy照射后P65P50明显增高,最高时超过假照组的200%(2~8h),而P50/P50则升高不明显;2Gy照射后则显示P65/P50、P50/P50均明显升高,二者在2~8h也超过假照组的200%.结论 IL-12表达增强和NF-κB活性升高的一致性提示NF-κB可能参与辐射诱导IL-12变化的分子调控。 |

Objective To observe the changes of IL-12 and NF -κB in murine peritoneal macrophages after whole-body irradiation(WBI) with different doses X-rays. Methods Enzyme-linked immunosorbent assay(ELISA) and electrophoresis mobility shift assay(EMSA) were used to detect the secretion of IL-12 and the DNA-binding activity of NF -κB, respectively. Results ELISA data indicated both 0 075 Gy and 2 Gy increased the secretion of IL-12 and the latter seemed to increase IL-12 more greater than the former.EMSA showed two bands representing P65/P50 and P50/P50 respectively. 0 075 Gy increased the P65/P50 activity and showed little effect on P50/P50, while 2 Gy increased both the two dimmers, consequently elevated the DNA-binding activity of NF -κB more significantly. Conclusion The results suggested that NF -κB might participate in the radiation-induced changes of IL-12. |
