      目的 探讨5Gy全身照射对伤口成纤维细胞(fibroblasts,FBs)功能的抑制效应及W11a12的促愈作用。方法 采用60Coγ射线5Gy全身照射大鼠复合背部植入聚乙烯醇海绵的皮肤伤口模型,观察伤后伤口FBs的纤维连接蛋白(FN)、转化生长因子β1(TGFβ1)、碱性成纤维细胞生长因子(bFGF)免疫细胞化学染色反应的变化及W11a12对其的影响。结果 5Gy全身照射复合皮肤创伤后第3、5天伤口内FN、TGFβ1、bFGF免疫细胞化学染色反应阳性的FBs数量均显着减少,但减少的程度有所不同,由重到轻依次为bFGF、FN、TGFβ1.而W11a12能显着增加未照射组和照射组伤口中FN、TGFβ1、bFGF免疫细胞化学染色反应阳性的FBs数量。结论 全身放射损伤对伤口中FBs的合成与分泌功能具有抑制作用,增强FBs合成和分泌功能是W11a12促愈作用的一个途径。

      Objective To investigate the inhibitory effects of 5 Gy whole body irradiation on wound fibroblast function and the promoting role of W11-a12 on these cells.Methods Two full thickness skin incision wounds,in which polyvinyl alcohol sponges were planted,were inflicted on both sides of the back of rats after they had received 5 Gy gamma ray whole body irradiation.The percentages of immunocytochemistry positive fibroblasts for fibronectin (FN),transforming growth factor β1(TGFβ1) and basic fibroblasts growth factor (bFGF) in the wound were observed.Results The percentages of immunocytochemistry positive fibroblasts for FN,TGFβ1 and bFGF in wound were all significantly reduced on day 3 and 5 after 5 Gy whole body irradiation,but the degree of reduction varied in severity (bFGF>FN>TGFβ1).W11-a12 could increase the percentages of immunopositive cells for FN,TGFβ1 and bFGF both in simple wound and in wound impaired by irradiation.Conclusion Whole body irradiation injury could cause significant inhibitory effect on wound fibroblast function of synthesizing and secreting FN,TGFβ1 and bFGF.W11-a12 could promote irradiation impaired wound healing by means of enhancing these functions of wound fibroblasts.
