目的 观察75mGy、2GyX射线全身照射对小鼠脾淋巴细胞中IL-10的影响。方法采用Northernblot检测IL-10转录水平的变化, 并同时通过流式细胞术检测其蛋白合成的变化。结果①蛋白合成结果表明, 75mGyX射线全身照射后脾淋巴细胞IL-10合成在照后2h开始即呈时间依赖性降低, 至48h仍无恢复迹象(P<0.01),而2Gy照射后则出现IL-10合成升高, 于照射后8h达到峰值(P<0.05).②Northernblot结果表明, 75mGy照射后脾细胞IL-10mRNA转录水平从照后1h即降低, 并维持较低水平一直到48h开始恢复, 达到假照射组的8835%;2GyX射线全身照射后早期mRNA水平即有所升高, 2h达到假照组的13406%,4h略有下降, 随后逐渐恢复至假照射组水平。结论 不同剂量X射线全身照射可引起不同的辐射效应, IL-10在其中起着重要的作用。 |

Objective To observe the time course changes in the expression of IL-10 in splenocytes after whole body irradiation (WBI) with different doses of X-rays. Methods Northern blot and flow cytometry(FCM) were adopted to detect the changes of IL-10 at mRNA and protein levels respectively. Results FCM data indicated IL-10 protein synthesis decreased beginning from 2 h after exposure and remained at low level up to 48 h after 75 mGy irradiation.The increase of IL-10 synthesis was detectable after 2 Gy irradiation.By northern blot we found IL-10 mRNA decreased significantly after WBI with 75 mGy X-rays(low dose radiation,LDR),while it increased at 2 h after WBI with 2 Gy X-rays (high dose radiation,HDR) . Conclusion The results suggest LDR and HDR have different effects on IL-10. |
