      目的 观察IL-2、4、6、7和IFN-γ对γ射线诱导的人扁桃体淋巴细胞凋亡的调节作用.方法 取手术切除的扁桃体组织,经Ficoll分离得单个核细胞,采用 4Gy60Coγ射线照射,用凝胶电泳显示细胞凋亡;体外用IL-2、IL-4、IL-6、IL-7及IFN-γ与单核细胞共培养 2 4h后流式细胞仪测CD4+ 、CD8+ T细胞及CD19+ B细胞的凋亡率及表面bcl-2的表达.结果 照射后 4~24h有明显的梯形条带出现;CD8+ T细胞和B细胞的辐射敏感性较CD4+T细胞高;IL-2、IL-4和IL-7对CD4+ 、CD8+T细胞凋亡及IL-4对B细胞凋亡有明显的保护作用,同时可观察到bcl-2明显的高表达.IL-6和IFN-γ无此作用.结论 细胞因子IL-2、IL-4与IL-7可促进放射病病人免疫功能恢复.

      Objective To observe the protective action of IL-2,4,6,7 and IFN-γ on radiation-induced apoptosis of human tonsil lymphocyte subpopulation. Methods Human tonsil specimens were obtained from routine tonsillectomies.Lymphocytes were separated and co-cultured with IL-2,4,6,7 and IFN-γ for 24 h,then exposed to 4 Gy γ-rays.DNA gel electrophoresis and flow cytometric light scatter analysis were performed to discriminated the viable cells from dead cells,and bcl-2 expressed on it. Results The "ladder pattern" of DNA fragmentation could be seen within 4-24 h after 4 Gy irradiation.CD8+T and B cells showed higher radiosensitivity than that of CD4 + T cells.The radiation-induced apoptosis in both CD4+ and CD8+ T cells was significantly inhibited and bcl-2 expression was induced by IL-2,IL-4 and IL-7.The viability of B cells was maintained and bcl-2 was overexpressed by the presence of IL-4 but not by others in culture. Conclusion We conclude that the protective effect by each cytokine on radiation-induced apoptosis might be partly attributed to their enhancement of cellular expression of bcl-2 protein.
