      目的 研究白介素6(IL6)在60Coγ辐射诱导人多发性骨髓瘤细胞株XG6-XG-7凋亡效应中的作用及其可能的分子生物学机理。方法 运用流式细胞仪技术(FACS)、DNA片段释放法和RT-PCR,观察了IL6对细胞株XG6、XG7中BclxL、BaxmRNA相对表达的影响及其与辐照诱导的细胞凋亡率的相关性。结果 IL-6存在时,XG6、XG7细胞的Bcl-xL、BaxmRNA表达在辐照后迅速上升,4h达最高峰后逐渐下降,24~48h恢复到照射前水平;不加IL6的XG6、XG7细胞的BclxL的表达在同样实验条件未检测到,而这2株细胞BaxmRNA的表达在照射后不同时间均低于相应的加IL6组,但尚有明显的表达。对不加IL6的细胞组,辐照后不同时间的凋亡率明显高于相应的加IL6组(P<0.05).结论 IL-6可通过调节BclxL、Bax的表达及改变BcL-xL/Bax二者比率,保护辐照诱导XG6、XG7的细胞凋亡。

      Objective To investigate interleukin 6 involvement in the response of multiple myeloma cell lines XG-6 and XG-7 to 60 Coγ-radiation. Methods Apoptosis was confirmed by flow cytometry and quantitated by the assay of DNA fragmentation release.The expression of Bcl-xL and Bax in XG-6 and XG-7 cells after irradiation and cultured with or without interleukin 6 was monitored by RT-PCR. Results XG-6 and XG-7 cells in the presence or absence of interleukin 6 underwent apoptosis at 24 h after γ-irradiation,but in the absence of interleukin 6 they had higher rates of γ-radiation-induced apoptosis than in the presence of interleukin 6(P<0.05) and had lower levels of Bcl-xL and Bax expression. Conclusion Interleukin 6 affects the sensitivity of the cell lines to γ-radiation-induced apoptosis and can rescue cells from apoptosis by up-regulating the expression of Bcl-xL and changing the Bcl-xL/Bax ratio.
