      目的 探讨血小板第4因子(PF4)对小鼠7.5Gyγ射线全身照射所致急性放射损伤的防护作用及机理。方法 小鼠照射前20h于腹腔内注射PF4两次,间隔6h,每次剂量20~60μg/kg.观察动物的30d存活情况及几种造血指标。结果 30d存活率对照组和防护组分别为0%和50%~60%(P<0.01);防护组的CFUGM集落形成数、骨髓细胞DNA含量、骨髓有核细胞数、脾结节形成数、脾体比、胸体比等均较照射对照组增加,经统计学处理差异有显着性(P<0.05)或有非常显着性(P<0.01).结论 PF4对小鼠急性放射损伤有防护作用,其机理可能同它保护和促进造血功能恢复有关。
英文摘要:
      Objective To investigate the protective effect and mechanism of platelet factor 4(PF4)on irradiated mice. Methods One hundred male mice were randomized to several groups and exposed whole-bodily to 7 5 Gy 60Co γ rays.The mice were treated with injections of PF4(20 μg/kg to 60 μg/kg)twice at an interval of 6 h and 20 h, after the second injection they were irradiated.The 30-day survival rated were recorded. Results The 30-day survival rate and the mean survival days, bone marrow cellularity.DNA content of bone marrow cells, number of CFU-GM on agar culture, peripheral blood cells, spleen/body weight ratio and number of spleen colonie were all significantly higher in the PF4 treated groups than in the corresponding irradiated controls (P<0.01). Conclusion PF4 has beneficial protective effect on irradiated mice.Its mechanism could be relatied to protection of hematopoietic cells and accele ration of their recovery.
