      目的 从伤口早期中性粒细胞(Neu)肌动蛋白(actin)的变化方面探讨全身γ射线照射后Neu吞噬、趋化功能变化的机理及促愈药康复新(W11-a12)的作用机理。方法 以大鼠6Gy全身照射复合皮肤切割伤为模型,收集伤口细胞。用Fluorescein鬼笔环肽标记Neu F-actin进行测定和观察。结果 复合伤组Neu F-actin含量先高后低,对FMLP反应性减弱,FMLP刺激后解聚慢,康复新能改善Neu F-actin的功能状态。结论 电离辐射引起Neu肌动蛋白功能异常可能是Neu趋化、吞噬活性降低的重要原因,改善Neu F-actin的功能状态是康复新提高Neu吞噬和趋化功能的作用机理。
英文摘要:
      Objective To explore the mechanism of decreased phagocyotic and chemotactic function of neutrophils (Neu) and the mechanism of W11-a12' s pharmacological action by studying the changes of actin of Neu in wound after total body irradiation of rats. Methods Cells were collected from incision wounds on back of rats after 6 Gy whole body-irradiation.F-actin in Neu was stained with Fluorescein-phalloidin. Results There were increased (3~12 h) and decreased (24 h, 48 h) basal levels of F-actin, a lower responsiveness to stimulation and a reduced rate of actin depolymerization after combined radiation injury compared with simple wound group, which were important causes of Neu dysfunction. Conclusion W11-a12 could improve the functional state of Neu actin, which may contribute to the improved chemotactic and phagocytotic function of Neu after administration of this drug.
