目的 探讨肠型放射病分子层次发生机理,寻找肠上皮细胞辐射损伤相关基因。方法 用mRNA差异显示技术比较研究12Gyγ射线辐射损伤前后小鼠小肠上皮细胞间基因表达的异同,分离并克隆差异表达基因片段,斑点杂交验证其表达特异性,对有意义的片段进行测序和同源性分析。结果 成功地克隆到一条辐射损伤特异表达基因片段,其与大鼠粘蛋白有部分同源(EMBL,57.456%).结论 mRNA差异显示技术能充分展示辐射损伤前后肠上皮细胞间基因表达的差异,通过反复的筛选和功能鉴定有可能揭示辐射引起肠上皮细胞损伤与修复的分子机理。 |

Objective To find genes differentially expressed in intestinal epithelial cells (IECs) induced by radiation so as to elucidate the molecular mechanism of intestinal type of radiation sickness. Methods mRNA differential display was used to study the difference of expressed genes in IESs in normal and irradiated mice 3h after exposure to 12 Gy γ-rays,then differentially expressed gene was cloned and dot blot hybridization was carried out to confirm its identification. Results A differentially expressed gene induced by radiation was successfully cloned. Conclusion mRNA differential display can fully reflect the difference in expressed genes between normal and radiation injured IECs.Therefore,large-scale screening and identification of differentially expressed genes are conducive to the revealment of the molecular mechanism of IECs damage due to ionizing radiation. |
