采用3H-TTP掺入法和DNA聚合酶α、γ选择性抑制剂N-乙基马来酰亚胺,研究了不同剂量、不同剂量率γ射线照射对重组大鼠DNA聚合酶β、BALB/c裸鼠肝细胞和SMMC-LTNM肝癌细胞核内DNA聚合酶β活力的影响。结果表明:受照离体肝细胞和肝癌细胞核内DNA聚合酶β活力无显着改变,重组大鼠DNA聚合酶β在受照40Gy时酶活力比未照组降低15%左右(P<0.05);小鼠整体受照2Gy后,上述两种细胞核内DNA聚合酶β活力在24~28小时内略有下降,随之出现一过性增高,呈现双相变化过程。 |

The effect of γ-rays on rodent DNA polymerase βactivity was studied with 3H-TTP incorporation inhibition test, using Nethylmaleimide as the selective inhibitor of DNA polymerase αand γ.The DNA polymerase βtested here included recombinant rat enzyme,enzyme in nuclei of normal hepatocytes and SMMC-LTNM hepatoma from BALB/c nude mice.It was found that the rat recombinant polymerase βactivity decreased markedly as the absorbed dose increased to over 40Gy.The enzyme activity in both kinds of nuclei was not significantly affected after exposure to γ-rays in vitro, whereas it decreased slightly within 24-48 hours after total body irradiation with 2 Gy of γ-rays, and then temporarily increased to over normal level. |
